Another question: using permission is there the possibility to select a direct access to the regioin timeline avoiding the possibility to modify/add region on the layout?

No, if you want user to have access to the designer he will need edit permissions to that layout and with that he will be able to add new regions (you can restrict access to existing regions - they can have separate permissions, but he will be able to create new regions)
