We utilise multiple playlists across many layouts and we constantly are adding and removing media from the playlists. The layouts do not automatically update when media is added or removed from the playlists. Do I have to manually re-publish all of the layouts (very impractical) or is there a way to auto update/re-publish the layouts?

Playlists are created and managed independently to Layouts so any changes made to a Playlist does not require you to make any changes to the Layout the Playlist is in. If the Layout is in a Published state any changes should be reflected the next time the Layout reloads or from what is set for the collect interval in your Display Settings.
